Selective: Bile inhibits the growth of most Gram-positive bacteria except for enterococci and some streptococci.
Differential: Esculin is hydrolyzed by certain bacteria, including enterococci, producing esculetin and glucose. Esculetin reacts with ferric citrate in the medium to form a dark brown or black complex.
Used for isolating and differentiating Enterococcus species and other group D streptococci.
Esculin hydrolysis is indicated by a black or brown coloration around the colonies.
Dissolve the appropriate amount of powder (as indicated on the product label, typically ~64 g/L) in distilled water.
Heat until completely dissolved and sterilize by autoclaving at 121°C for 15 minutes.
